10th December 2025 - In section 1, 'Overview of the protocol', in the Table under 'Library preparation', timing for the flow cell priming and loading was increased from 5 min to 10 min. And in 'Compatibility of this protocol', MinION Mk1B and MinION Mk1C along with their links to the IT requirements documents were removed as these documents have moved to legacy. - In sections 3 and 6 for the device branches, the link for the flow cell check was updated. - In section 4, an important note was included stating that if using the barcode primers in plate format, to only use the wells once. - In section 6, 'Priming and loading the MinION/GridION Flow Cell', a tip was added to bring the flow cell to room temperature for better visibility of the array. And the addition of BSA to the flow cell priming mix was changed from a 'recommended' to a 'required' instruction. A note was added to the end of this section to explain that when a flow cell is inserted into the MinION Mk1D, there will be a small gap around the sides of the device. - In section 7, 'Data acquisition and basecalling', links for the MinION Mk1B and MinION Mk1C user manuals were removed as these documents have moved to legacy. And in 'To start a sequencing run in MinKNOW', point 3, the kit used for sequencing was specified for the kit selection page on MinKNOW. - In 'Troubleshooting', section 11, Guppy was removed as it is no longer applicable.
