Telomere-to-telomere phased genome assembly using error-corrected Simplex nanopore reads

Three horizontal parallel strands of DNA with the bases in blue, yellow and green with the letters floating above. Each strand has the same order of bases

For telomere-to-telomere studies, many researchers use at least two long-read sequencing technologies. This approach results in increased costs and a requirement for a higher starting amount of DNA. Here the authors have developed the HERRO model which error corrects nanopore simplex ultra-long reads, increasing accuracy up to 100-fold. This data can then be used to achieve highly contiguous assemblies comparable to a combined sequencing approach.
