24th November 2025 - In section 1, in the Table under 'Library preparation', the timing for the flow cell priming and loading was changed from 5 min to 10 min and the workflow image was updated to reflect this change. In 'Compatibility of this protocol' MinION Mk1B and MinION Mk1C were removed along with their links for IT requirements as these documents have moved to legacy. This also includes removal of links to their user manuals in 'Data acquisition and basecalling'. - In section 3, 'Library preparation', check your flow cell, the link for the flow cell check was edited. And an important note was added to only use the barcode wells once. - In section 4, 'Flongle Flow Cell loading', the video link for flow cell loading was updated. - In section 4, 'Priming and loading the MinION/GridION Flow Cell', a tip was added to bring the flow cell to room temperature for better visibility of the array. And the addition of BSA to the flow cell priming mix was changed from a 'recommended' to a 'required' instruction. A note was added to the end of this section and for the Flongle Flow Cell loading to explain that when a flow cell is inserted into the MinION Mk1D, there will be a small gap around the sides of the device. - In section 5, 'Data acquisition and basecalling', screenshots were removed for the Flongle device branch and the name of the sequencing kit was specified for the kit selection page in MinKNOW for all device branches.
