- In 'Overview of the protocol', links for the EPI2ME amplicon workflow were updated. And in 'Library preparation', the text 'Adapter ligation' was updated to 'Rapid adapter attachment' to signify rapid chemistry and the timing for priming and loading a flow cell was increased from 5 min to 10 min in the Table.
- In 'Compatibility of this protocol', MinION Mk1B and Mk1C were removed, along with their links for IT requirements as these device documents have moved to legacy. This also includes removal of links to their user manuals in 'Data acquisition and basecalling'.
- In section 2, 'Equipment and consumables' text in the SQK-RBK114.96 kit image was edited.
- In section 3, 'Library preparation' , 'Check your flow cell' and section 4, 'Priming and loading the flow cell', 'Complete a flow cell check', the links for the flow cell check in the MinKNOW protocol were replaced with links for the flow cell check document. And in section 3, a note was added to use the barcode wells only once.
- In section 4, 'Priming and loading the MinION/GridION Flow Cell', a tip was added to bring the flow cell to room temperature for better visibility of the array. And the addition of BSA to the flow cell priming mix was changed from a 'recommended' to a 'required' instruction. A note was added to the end of this section to explain that when a flow cell is inserted into the MinION Mk1D, there will be a small gap around the sides of the device.
- In 'Data acquisition and basecalling', screenshots for MinKNOW were removed.
- In 'Troubleshooting', sections on Voltrax and Kit 9 chemistry were removed as they are no longer supported.
