1. What is UBik?
What is UBik?
UBik is a sample delivery device to enable sequencing directly from a sample, removing the need for complex sample preparation and allowing the non-skilled user to obtain sequencing information from our technology. All reagents are contained within the device and/or flow cell.
The input can be from a range of biological or environmental samples e.g. saliva, blood, bacterial culture, urine, swab sticks.
UBik replaces the need for a library preparation kit and operates with Flongle and MinION Mk1B or GridION.
This device is aimed at the non-skilled user with no infrastructure e.g. home user, in-field scientist or nosocomial environments.

How it works
The device is simple to use (Figure 1.):

Sample is added to the UBik by undoing a screw cap. Once your sample is added, the cells undergo lysis releasing DNA. Manual shaking of the device agitates the large beads within the chamber, resulting in efficient dispersal of the sample and mixing of the reagents. Cellular proteins, including DNAses, are also denatured in this step. DNA is captured on small beads, which collect at the bottom of the tube.
The twist-off seal is broken, and the open port of the delivery device can be docked onto the Flongle flow cell to release the DNA-bound beads.
Once the bead-bound DNA is delivered into the flow cell, the DNA is released from the beads, and adapters are added to the DNA. The adapters create compatible ends for our sequencing chemistry, and are pre-loaded with the motor protein that ratchets DNA through the pore. Adapters and pores are identical to those used in the standard library preparation kits.
