- Protocol was updated to align with current formatting.
- In section 1, 'Steps in the sequencing overflow', under 'Library preparation', the timing for priming and loading a flow cell was increased from 5 min to 10 min in the Table. And under 'Sequencing', Guppy was replaced with Dorado as an optional software for demultiplexing as Guppy is no longer supported.
- In section 4, 'Native barcode ligation', an important note was included to only use the barcode wells once.
- In section 5, 'Adapter ligation and clean-up', and section 6, 'Priming and loading multiple flow cells', a note was added to check your flow cell or complete a flow cell check.
- In 'Troubleshooting', video links for the MinION Flow Cell loading were replaced with the PromethION Flow Cell loading.
