Comparison of DNA extraction methods for nanopore sequencing of complex plant and animal genomes

Over the past decade, long-read, single-molecule DNA sequencing technologies have emerged as influential players in the genomic arena. In this regard, the isolation of high-quality and high-molecular-weight DNA is needed to fully take advantage of the potential of these third-generation technologies. Such steps become challenging, particularly when dealing with complex genomes of plants or animals. Here, we performed an in-depth analysis of a large number of commercial and in-house-adapted gDNA extraction methods to assess their relative performance across a large variety of samples: plant, insect, and two scleractinian corals.
