These protocols describe methods for DNA extraction from a variety of rabbit and rat tissues as well as from bull sperm, as examples of mammalian samples. We found that the throughput obtained from samples extracted from blood (collected in K2-EDTA) typically generated highest sequencing output and long read lengths. We recommend the use of SPRI size selection prior to sequencing. The sequencing performance was assessed using the Ligation Sequencing Kit.
