DNA extraction from dried blood spots for Oxford Nanopore sequencing

Poster for ACMG by Tilley et al.

This poster presents an optimised workflow for extracting high molecular-weight (HMW) DNA from dried blood spots (DBS) for whole-genome sequencing (WGS) and Adaptive Sampling (AS) using Oxford Nanopore technology. By evaluating multiple DBS card types and working with inputs below recommended levels, we demonstrate that DBS-derived DNA can deliver high-quality long-read data suitable for variant detection, methylation analysis, and targeted enrichment, even from limited input material.

  • How a robust HMW DNA extraction protocol enables long-read nanopore WGS that is compatible with multiple different DBS card types, even from limited inputs
  • How AS increases on-target coverage (80–100×) of a pharmacogenomic (PGx) panel while retaining genome-wide coverage (>10× outside targeted regions), enabling combined targeted and low-pass analyses
  • A single-platform approach that enables comprehensive genomic and epigenomic analysis of DBS samples
