10th December 2025 - In section 1, 'Overview of the protocol', under subsection 'Steps in the sequencing workflow' and in section 10, 'Downstream analysis', users are advised to use the Dorado basecaller for analysis. And in 'Compatibility of this protocol', MinION Mk1B and Mk1C were removed, along with their links for IT requirements as these device documents have moved to legacy. This also includes removal of links to their user manuals in 'Data acquisition and basecalling'. - In section 2, 'Equipment and consumables', single-use tube format for the SQK-LSK114 kit was removed as this format is no longer applicable. - In section 5, 'cDNA repair and end-prep', 'Check your flow cell' and section 7, 'Priming and loading the flow cell', 'Complete a flow cell check', the link for the flow cell check in the MinKNOW protocol was replaced with the link for the flow cell check document. - In section 7, 'Priming and loading the MinION and GridION Flow Cell', a tip was added to bring the flow cell to room temperature for better visibility of the array. The video link for 'Priming and loading your flow cell' was updated and in step 2, for all device branches, the instruction for using the single-use tube format for the LSK114 kit was removed. The addition of BSA to the flow cell priming mix was changed from a 'recommended' to a 'required' instruction. And an important note was added to the bottom of this section for MinION stating that when a flow cell is inserted into the MinION Mk1D and the lid is closed, there will be a small gap around the sides of the device. - Section 8, 'Data acquisition and basecalling' was formatted to align with current protocols.
