These protocols describe the methods used to extract gDNA from a variety of plant leaf samples. Extracted DNA is size selected using the size selection of HMW DNA by semi-selective DNA precipitation protocol and sequenced using the Ligation Sequencing Kit. We recommend extracting the DNA with Carlson lysis buffer and purifying using the QIAGEN Genomic-tip 500/G.
