Evaluation of Oxford Nanopore whole-genome methylation sequencing for phenotype inference and association testing

  • Published on:
Scientific poster titled "Evaluation of Oxford Nanopore whole genome methylation sequencing." Includes graphs and text.

This poster evaluates Oxford Nanopore whole-genome methylation sequencing (WGMS) across three common methylation applications: epigenetic age prediction ('epigenetic clocks'), whole-blood cell-type deconvolution, and a proof-of-principle epigenome-wide association study (EWAS). Using healthy whole-blood samples with matched EPIC array data, we show how WGMS compares with arrays while reaching CpG sites that arrays do not target.

Download this poster to discover:

  • How WGMS methylation estimates closely match those from EPIC v2 arrays at shared CpG sites
  • How WGMS performed comparably to arrays for epigenetic age prediction, cell-type deconvolution, and EWAS
  • Why these results suggest WGMS can serve as a direct replacement for array-based methylation analyses
  • How WGMS extends coverage to the large majority of CpGs that arrays miss
  • How sequence and haplotype context is added alongside methylation data
