Cover Your Bases: How to Minimize the Sequencing Coverage in DNA Storage Systems

This study investigates the minimal coverage required for accurate reconstruction of synthetic DNA. By analysing the effects of read quality, DNA library design, and reconstruction algorithms, we optimize the sequencing process and thereby reduce the number of reads required. By exploiting the cutting-edge technology of ONT MinION, which allows for real-time sequencing and direct reconstruction, the reduction in coverage leads to reduced latency and costs.
