De novo whole-genome assembly of Chrysanthemum Makinoi


Although Chrysanthemum represents one of the most economically important ornamental crops, genomes have only been partially assembled for 2 diploids, and the cultivated hexaploid genome is still poorly understood. We present a novel assembly for a key wild diploid based on long-read sequencing technology with short-read polishing. With a total length of 3.113 Gb, N50 of 330.01 Mb and BUSCO complete score of 92.1%, we have created the most contiguous Chrysanthemum assembly to date. This assembly provides further insight into the origin of the cultivated hexaploid as well as the most effective strategies to sequence and assemble the polyploid varieties.
