Comprehensive characterisation of the mouse retina transcriptome using long-read RNA sequencing

In this webinar, Rui Chen evaluates Oxford Nanopore RNA sequencing technology for transcriptome profiling at the single-cell level, and for the generation of a comprehensive atlas of full-length transcript isoforms in the mouse retina. Chen’s team profiled approximately 30,000 mouse retina cells using a combination of 1.4 billion long reads with nanopore sequencing and 1.5 billion short reads with legacy sequencing.

  • Nanopore long-read scRNA-seq is highly effective for the identification of isoforms

  • Isoforms exhibit differential expression across cell class and cell subclass

  • Although rare, over 2,000 cell class specific isoforms were identified by the lab, with potential use going forward as a cell class marker.
