Extraction Method Group
Avian blood gDNA
These protocols describe two methods for DNA extraction from chicken blood, as an example of avian blood. We recommend the QIAGEN Genomic-tip 100/G (7.5 µl of blood) and the QIAamp DNA Blood Maxi Kit (200 µl of blood): the Genomic-tip method yielded long read lengths and moderate sequencing throughput, and while higher recovery of DNA was observed with the QIAamp DNA Blood Maxi Kit (note that this is with a larger sample input), much shorter read lengths were observed. We also recommend the use of SPRI size selection prior to sequencing. To obtain more sequencing data from this sample, users may find that their experiments would benefit from a flow cell wash step.
Please see below a comparison of DNA fragment lengths, sequencing read lengths, and throughput for the five protocols. Click on the panel of your choice to view the protocol.
