Polyadenylation is an essential modification to nearly all RNAs, playing a pivotal role in the control of the transcript’s stability. Herein, we used direct RNA sequencing to analyze poly(A) tails in yeast Saccharomyces cerevisiae devoid of enzymes relevant for poly(A) tail metabolism. Our comprehensive sequencing data supported by validation experiments confirmed many previously suggested trends regarding poly(A) tails but also challenged the established knowledge in the field. It also provides a large resource of direct RNA sequencing data which can be related to other crucial aspects of post-transcriptional gene expression regulatory mechanisms in budding yeast.
