Targeted nanopore sequencing with Cas9-guided adapter ligation
In this publication, Gilpatrick et al. describe their nanopore Cas9-Targeted Sequencing (nCATS) method. Focusing on 10 cancer genes, they used this technique to interrogate methylation and detect variants in cancer cell lines and tumour tissue samples. Long nanopore sequencing reads facilitated the identification of structural variants at target loci, and simplified the phasing of methylation into different alleles, allowing the team to investigate allele-specific epigenetic changes.
